چکیده

Atopic dermatitis (AD) is characterized by skin barrier dysfunction, increased type 2 immunity, and an altered microbiota with high Staphylococcus aureus abundance. These factors contribute to enhanced susceptibility viral infections. We hypothesize that both immunity (IL-4/13) S. colonization alter the epithelium make it highly permissive Using human keratinocytes (KCs), we observed state of differentiation significantly affected vaccinia virus (VV). Similar trends were herpes simplex virus. Undifferentiated KCs relatively resistant VV infection, whereas infection at time or up days later 13- 6-fold, respectively (% area plaques; n=5-9, p<0.001). This highlights a narrow window in which are susceptible implicates role for tight junctions (TJ) KC infectivity. found exposed USA300 supernatant IL-4/13 (50 ng/mL) had (7 & 8-fold increases plaque number; p<0.01 p<0.05 respectively) day post-differentiation. To further explore this observation infectivity associated changes differentiation, determined whether these stimuli effect on TJ function (Transepithelial Electrical Resistance [TEER]). induced decrease TEER (>80%, p<0.02) reduced accumulation protein, occludin (p<0.01). The variable. currently exploring expression markers using laser capture full thickness epidermal explants identity strata support replication. findings will clarify layers (differentiation states) most infections modified exposure virulence and/or cytokines.

Barrier in Epidermal Keratinocytes Activation of TLR2 Enhances Tight Junction

The epidermis has developed physical and immunological barriers that prevent infiltration of deleterious chemicals and pathogens. As a first step to understanding the relationship between these barriers, we investigated whether TLR2 activation functionally alters tight junctions (TJs) in cultured human keratinocytes. Stimulation with peptidoglycan, a ligand for TLR2, elevated the TJ-associated ...

Dynamic changes in murine forebrain miR-211 expression associate with cholinergic imbalances and epileptiform activity.

Epilepsy is a common neurological disease, manifested in unprovoked recurrent seizures. Epileptogenesis may develop due to genetic or pharmacological origins or following injury, but it remains unclear how the unaffected brain escapes this susceptibility to seizures. Here, we report that dynamic changes in forebrain microRNA (miR)-211 in the mouse brain shift the threshold for spontaneous and p...

Tight junction proteins form large complexes and associate with the cytoskeleton in an ATP depletion model for reversible junction assembly.

A key feature of the ischemic epithelial cell phenotype is the disruption of tight junctions (TJ). In a Manin-Darby canine kidney cell model for ischemia-reperfusion/hypoxia-reoxygenation injury which employs inhibitors of glycolysis (2-deoxy-D-glucose) and oxidative phosphorylation (antimycin A), transepithelial electrical resistance, a measure of TJ integrity, dropped rapidly, correlating wel...

External antigen uptake by Langerhans cells with reorganization of epidermal tight junction barriers

Outermost barriers are critical for terrestrial animals to avoid desiccation and to protect their bodies from foreign insults. Mammalian skin consists of two sets of barriers: stratum corneum (SC) and tight junctions (TJs). How acquisition of external antigens (Ags) by epidermal Langerhans cells (LCs) occur despite these barriers has remained unknown. We show that activation-induced LCs elongat...
